The Trellis team, embedded in the Departments of ‘Cellular and Molecular Medicine’ and ‘Microbiology, Immunology and Transplantation’ at KU Leuven, is looking for a motivated and skilled research expert to join its Gene Therapy Discovery Science team and to conduct research on several aspects of adeno-associated virus (AAV)-based gene therapy. The senior researcher will act as an expert in AAV viral vector production (upstream), analytical development and capsid engineering.

WHERE WILL YOU BE WORKING?
Trellis, headed by prof. Els Henckaerts, is a pioneering lab that works on the cutting edge of AAV biology research and translational science to develop novel AAV-based gene therapies for the clinic.
Recent advances in gene therapy have boosted the commercial value of clinical AAV vectors. However, large-scale manufacture of high-quality vectors remains a challenge. So, the need for innovative solutions in upstream and downstream bioprocessing is indisputable. Exploring and implementing innovative technologies and processes will be part of your job. This allows you to influence and drive the long-term ambition to develop a scalable and robust manufacturing platform for AAV vectors within KU Leuven.

Responsibilities

  • Upstream development of a scalable and robust manufacturing platform for AAV vectors.
  • Applying bioprocess engineering strategies to optimize AAV production in suspension cells. Knowledge of DOE design and power analysis is a plus.
  • Independently planning and executing studies on the development of the manufacturing process and analytical testing of AAV viral vectors.
  • Exploring, stimulating and implementing innovative technologies and processes in the field.
  • Designing, executing and documenting assays to characterize and quantify viral vectors, including managing collaborations with partners and external service providers.
  • Contributing to novel AAV capsid and vector engineering efforts to tackle current challenges in the field.
  • Managing a team of 2-4 technicians, and providing guidance to M.Sc. and Ph.D. students.
  • Applying expertise in manufacturing and analytical testing to review workplans, activities and reports and maintain timelines and budgets.
  • Establishing and nurturing collaborations with national and international academic and industrial partners on gene therapy product development and production.
  • Serving as a subject matter expert in AAV viral vector production, providing technical guidance and insights to the team and stakeholders.
  • Actively participating in internal and external research meetings to contribute insights and foster collaboration.

Profile

  • You hold a PhD degree in Virology, Biochemistry, Cell Biology, Molecular Biology, Biomedical Engineering, or related discipline 
  • You have at least 5 years hands-on experience in upstream process development for AAV manufacture in industry and/or academia.
  • You have deep expertise with AAV gene therapy vector production and analytical testing.
  • You have a solid understanding of the fundamentals of molecular biology and AAV-based gene therapy.
  • You are proficient in protein biochemistry, with proven hands-on experience in developing and executing biochemical and cell-based assays in high-throughput screening formats.
  • You have good communication skills, both written and oral, in English.
  • You have knowledge and experience in gene therapy and strong analytic thinking skills.
  • You have strong inter-personal and communication skills. Proven record of collaborations and effective work with academic and industrial partners. 
  • You possess strong organizational and managerial skills within a research context.
  • You have the ability to take initiative, work independently, and drive projects forward with minimal supervision
